The M100-3000 molecular weight DNA ladder is designed for determining the size of PCR products and dsDNA fragments in the 100-3000 bp range. The M100-3000 DNA Ladder is a mixture of specially selected, purified blunt-ended PCR products. The DNA Ladder gives a clear band pattern, with two bands of increased intensity employed as reference points. A DNA loading buffer (6x GREEN) is provided with each DNA ladder. The loading buffer contains Ficoll®400, which facilitates rapid sinking to the bottom of the well in the agarose or polyacrylamide gel. The solution protects DNA thanks to the presence of EDTA, which inactivates nucleases and other enzymes by binding the divalent metal ions crucial to their activity. The 6x GREEN solution contains two different dyes which migrate in the electrophoretic field differently from the standard size PCR products, ensuring that they do not hinder the analysis by image obscuration. The orange G dye corresponds to 50 bp and xylene cyanol FF to 4000 bp PCR products in a 1% agarose gel.

Storage Buffer (TE buffer)

10 mM Tris-HCl (pH 7.6) with 1 mM EDTA

6x Green DNA Loading Dye (supplied with the ladder)

20 mM Tris-HCl (pH 7.6), 0.03% xylene cyanol FF, 0.15% orange G, 15% Ficoll® 400 and 60 mM EDTA


- mix 1 volume of the dye with 5 volumes of the DNA solution

- do not heat before loading

- we recommend loading 10 µl of DNA ladder per gel lane

Quality Control

- tested in appropriate gel electrophoresis

- the absence of nucleases is confirmed by appropriate tests


Store at room temperature or at 4°C. For longer periods, store at -20°C (recommended).
