6x Green DNA Gel Loading Dye is pre-mixed loading buffer with a tracking dye for agarose and non-denaturing poylacrylamide gel electrophoresis. This solution contains EDTA that binds divalent metal ions, inhibits metal dependent enzymatic reactions and protects DNA from metal dependent nucleases, effectively inactivating enzymes and protecting DNA from nucleases. The loading dyes increase the density of the sample and they add colour to the sample, thereby simplifying the loading process. The optimized 6x Green DNA Loading Dye contains two electrophoresis tracking dyes: orange G and xylene cyanol FF. Orange G migrates at approximately 50 bp on a standard 1% TAE or TBE agarose gels, whereas xylene cyanol FF migrates at approx. 4 kbp.
Orange G dye allows for the monitoring of DNA migration on agarose and polyacrylamide gels; it migrates with the smallest DNA fragments without masking them under UV light. Because xylene cyanol FF migrates much slower than orange G, it is useful as a tracking dye when running gels for an extended time - long after orange G has run off the bottom of the gel.
6x Green DNA Gel Loading Dye
20 mM Tris-HCl (pH 7.6), 0.03% xylene cyanol FF, 0.15% orange G, 15% Ficoll® 400 and 60 mM EDTA
Use 1 µl of 6x Green Gel Loading Dye per 5 µl of DNA sample. Mix well before loading onto a gel.
Tested for DNA samplepreparation prior to agarose gel electrophoresis.
Store at room temperature or at +4°C up to 12 months. For longer periods, store at -20°C.
